使用方法如下：
1．将一侧目镜从镜筒中拔出，旋开目镜下面的部分，将目镜侧微尺刻度向下装在目镜的焦平面上，重新把旋下的部分装回目镜，然后把目镜插回镜筒中。
2．将镜台侧微尺刻度向上放在镜台上夹好，使侧微尺分度位于视野中央。调焦至能看清镜台侧微尺的分度。
3．小心移动镜台侧微尺和转动目镜侧微尺，使两尺左边的一直线重合，然后由左向右找出两尺另一次重合的直线。
4．纪录两条重合线间目镜侧微尺和镜台侧微尺的格数。计算目镜侧微尺每格所代表的实际长度。目镜尺每格所代表的实际长度=（两重合线间镜台测微尺的格数/两重合线间目镜测微尺的格数）×10um
5．取下镜台侧微尺，换上需要测量的玻片标本，用目镜侧微尺测量标本。
